Free radicals or “reactive oxygen species” may be initiated within cells by:

•Absorption of radiant energy such as UVLs and XRs

•Enzymatic metabolism of exogenous chemicals or drugs such as Carbon Tetrachloride

•Reduction-oxidation reactions that occur during normal metabolic processes. During inflammation, rapid bursts of Superoxide radicals can activate different polymorph nuclear leukocytes that cause further damage to skin cells

•Transition metals such as iron and copper can catalyze free-radical formations

•Nitric Oxide, an important chemical mediator generated by endothelial cells, macrophages, neurons and other cell types can also act as damaging free-radicals.

The effects of these reactive species are wide-ranging, but three reactions are particularly relevant to cell injury:

1.Lipid peroxidation of membranes. The lipid-radical interactions yield peroxides, which are themselves unstable and reactive, and an autocatalytic chain reaction ensues, which can result in extensive membrane, organellar, and cellular damage.

2.Oxidative modification of proteins. These changes accelerate degradation of critical enzymes by the multicatalytic proteasome complex, raising havoc throughout the cell.

3.Lesions in DNA. Reactions with thymine in nuclear and mitochondrial DNA produce single-stranded breaks in DNA. This DNA damage has been implicated in cell aging. Lingzshi polysaccharides and free radicals Lingzhi polysaccharides possess properties which can remove the free-radicals and help prevent related body damage by stopping the effects of these reactive species. Inactivation of free radicals includes the following properties of Lingzhi polysaccharides:

1.Antioxidants, which either block the initiation of free- radical formation or scavenge and terminate radical damage

2.Similarities to enzymes which act as free radical-scavenging systems and break down Hydrogen Peroxide and Superoxide anion.

These enzymes are:

(a) Catalase ,which decomposes Hydrogen Peroxide. Catalases are present in some bacteria --Staphylococcus Aureus for example, can cause acne breakouts

(b) Superoxide Dismutases, which can remove the free- radicals and help prevent related body damage by stopping hyper-oxidation of lipids. Such action protect the cells and delay the aging of cells

(c) Glutathione Peroxidase, which protects against injury by catalyzing free radical breakdown. The intracellular ratio of oxidized glutathione (GSSG) to reduced glutathione is a reflection of the oxidative state of the cell and is an important aspect of the cell's ability to detoxify reactive oxygen species.
